Objectives

After completing the course, students will be able to:perform qualitative and quantitative analysis of chromatographic data; estimate retention dependencies and select proper separation and detection conditions for analysis and purification of organic compounds; perform routine analysis with TLC, GC and HPLC and routine separation with preparative LC.

Core contentApplication area of modern chromathography. Separation principles. Analytical and preparative methods. Chromatography types and modes (TLC, LC, HPLC, GC). Stationary and mobile phases used for different analytes. Basic retention dependencies in GC, LC, SEC.Theory of chromatography. Van-Deemter equation.Chromatography instrumentation. Detectors for GC and HPLC. Qualitative and quantitative analysis. Retention parameters.Practice in qualitative analysis by TLC, preparative separation by LC, quantitative analysis by GC and HPLC.Complementary knowledgeAdvanced knowledge of retention dependencies.Separation efficiency dependencies.Choice of the optimal detection method for particular analyte based on selectivity and sensitivity.Specialist knowledgeOptimization of separation conditions.

Prerequisites

The prerequisites are Laboratory Safety course (ENS) or Basic laboratory course (MET), or a corresponding one from TAMK curriculum
